@#<b>Objective</b> To investigate the synergistic protective effects of WR-2721 combined with lentinan and cytokines against radiation damage in mice, and to provide a new treatment for acute radiation injury. <b>Methods</b> Seventy Institute of Cancer Research mice were divided into seven groups: a control group, a model group, WR-2721 group, Lentinan & cytokine group, WR-2721 & Lentinan group, WR-2721 & cytokine group and WR-2721 & Lentinan & cytokine group. All groups except the control group were irradiated with <sup>60</sup>Co γ-rays at a dose rate of 0.8 Gy/min and a cumulative dose of 5.0 Gy. The mice were sacrificed by cervical dislocation 14 d after irradiation to measure their spleen index, thymus index, and serum levels of superoxide dismutase (SOD), malondialdehyde (MDA), interleukin-11 (IL-11), and tumor necrosis factor-alpha (TNF-α). <b>Results</b> For the mice treated with WR-2721, lentinan, and cytokines, the spleen index was 7.33 ± 2.84, the thymus index was 1.70 ± 0.30, the serum SOD level was 114.0 ± 8.3, the MDA level was 7.33 ± 1.16, the IL-11 level was 155.8 ± 49.4, and the TNF-α level was 174.0 ± 37.8. All these indicators except the spleen index in the combination group significantly differed from those of the model group (<i>P</i> < 0.05 or 0.01), indicating the combined treatment promoted recovery from radiation damage. <b>Conclusion</b> WR-2721 combined with lentinan and cytokines has significant synergistic protective effects, which is a promising treatment for acute radiation injury.

RESUMO

An effective therapeutic regimen for hepatic fibrosis requires a deep understanding of the pathogenesis mechanism. Hepatic fibrosis is characterized by activated hepatic stellate cells (aHSCs) with an excessive production of extracellular matrix. Although promoted activation of HSCs by M2 macrophages has been demonstrated, the molecular mechanism involved remains ambiguous. Herein, we propose that the vitamin D receptor (VDR) involved in macrophage polarization may regulate the communication between macrophages and HSCs by changing the functions of exosomes. We confirm that activating the VDR can inhibit the effect of M2 macrophages on HSC activation. The exosomes derived from M2 macrophages can promote HSC activation, while stimulating VDR alters the protein profiles and reverses their roles in M2 macrophage exosomes. Smooth muscle cell-associated protein 5 (SMAP-5) was found to be the key effector protein in promoting HSC activation by regulating autophagy flux. Building on these results, we show that a combined treatment of a VDR agonist and a macrophage-targeted exosomal secretion inhibitor achieves an excellent anti-hepatic fibrosis effect. In this study, we aim to elucidate the association between VDR and macrophages in HSC activation. The results contribute to our understanding of the pathogenesis mechanism of hepatic fibrosis, and provide potential therapeutic targets for its treatment.

OBJECTIVE: To study the effects of cimetidine on low dose rate irradiation-induced liver cell apoptosis in Beagle dogs. METHODS: Healthy male Beagle dogs were randomly divided into normal control group, model control group, positive drug group (lentinan, 21.33 mg/kg) and cimetidine low-dose, medium-dose and high-dose groups (5.33, 10.67, 21.33 mg/kg), with 4 Beagle dogs each. Except for normal control group, other groups were given 60Co-γ accumulative irradiation (dosage rate: 0.040 8 mGy/min) for 23 d; the medication groups were given relevant medicine orally before irradiation, once a day. Twenty-four hours after stopping irradiation, TUNEL method was used to detect the apoptosis of liver cells in Beagle dogs. The percentage of apoptotic cells was calculated. The expression level of apoptosis-related proteins (Bax, Bcl-2, Caspase-3, p53) in liver tissue was detected by immunohistochemistry. RESULTS: Compared with normal control group, apoptotic cells and Bax, Caspase-3, p53 positive cells were increased significantly in liver tissue of Beagle dogs in model control group; the percentage of apoptotic cells, protein expression levels of Bax, Caspase-3 and p53 were increased significantly; Bcl-2 positive cells were decreased significantly, and its protein expression level was decreased significantly (P<0.05 or P<0.01). Compared with model control group, above positive cells of liver tissue in Beagle dogs were changed to different extents in medication groups; the percentage of apoptotic cells and protein expression levels of p53 in medication groups, protein expression levels of Bax in positive drug group, cimetidine low-dose and high-dose groups as well as protein expression levels of Caspase-3 in cimetidine groups were decreased significantly; protein expression levels of Bcl-2 were increased significantly in cimetidine groups. The percentage of apoptotic cells in cimetidine medium-dose and high-dose groups as well as protein expression levels of Caspase-3 in cimetidine groups were all lower than positive control group. Protein expression level of p53 in cimetidine low-dose group was significantly higher than positive drug group (P<0.05 or P<0.01). CONCLUSIONS: Cimetidine can inhibit the low dose rate irradiation-induced apoptosis of liver cells in Beagle dogs, and certainly protect liver cells against irradiation. The mechanism of it may be associated with up-regulating the protein expression of Bcl-2 and down-regulating the protein expression of Bax, Caspase-3 and p53 in liver cells.

RESUMO

Mueller polarimetry is a quantitative polarized light imaging modality that is capable of label-free visualization of tissue pathology, does not require extensive sample preparation, and is suitable for wide-field tissue analysis. It holds promise for selected applications in biomedicine, but polarimetry systems are often constrained by limited end-user accessibility and/or long-imaging times. In order to address these needs, we designed a multiscale-polarimetry module that easily couples to a commercially available stereo zoom microscope. This paper describes the module design and provides initial polarimetry imaging results from a murine preclinical breast cancer model and human breast cancer samples. The resultant polarimetry module has variable resolution and field of view, is low-cost, and is simple to switch in or out of a commercial microscope. The module can reduce long imaging times by adopting the main imaging approach used in pathology: scanning at low resolution to identify regions of interest, then at high resolution to inspect the regions in detail. Preliminary results show how the system can aid in region of interest identification for pathology, but also highlight that more work is needed to understand how tissue structures of pathological interest appear in Mueller polarimetry images across varying spatial zoom scales.

RESUMO

AIM To study the chemical constituents from Buthus martensii Karsch.METHODS The dichloromethane and 70% methanol fractions of B.martensii extract were isolated and purified by silica,ODS,Sephadex LH-20 and mid-pressure preparative column,then the structures of obtained compounds were identified by physiochemical properties and spectral data.RESULTS Nine compounds were isolated and identified as cholest4-en-3-one (1),cholesterol (2),uracil (3),1-stearyl-2-lyso-sn-glycero-3-phosphatidylcholine (4),glycerol (5),oleic acid (6),proline (7),alanine (8),leucine (9).CONCLUSION Compounds 1,4,5 are isolated from B.martensii for the first time.

RESUMO

Objective To investigate the radioprotective effect of cimetidine on survival rate and hematopoietic system in acutely irradiated mice.Methods The total body irradiation doses were 6.0Gy and 8.0Gy respectively at 1.01Gy/min rate. Sixty healthy male C57BL/6 mice were randomly divided into control group, model group, positive-drug (523) group and cimetidine groups (33.3mg/kg, 100mg/kg and 300mg/kg). Each group had ten mice. The mice were given intragastric administration of cimetidine for 6d before the irradiation in cimetidine groups, and 523 was administered before irradiation once a day for one day in 523 group, and at 5h after irradiation, was given again. The 30d survival rate after 8.0Gy irradiation was recorded. The peripheral blood cells, bone marrow DNA content and frequency of micronucleated polychromatic erythrocytes (fMNPCE) were determined 30d after 6.0Gy irradiation.Results After 8.0Gy irradiation, all the mice died on 21th day in model control group. The survival rates in cimetidine groups were 50%, 20% and 30%, respectively. After 6.0Gy irradiation on 30th day, compared with control group, the peripheral white blood cells (WBC) and bone marrow DNA content were decreased significantly (P<0.01,P<0.05) in model group, and fMNPCE was increased significantly (P<0.05). Compared with model group, WBC was significantly increased in 300mg/kg cimetidine group (P<0.01). In cimetidine groups, the bone marrow DNA content was increased significantly after irradiation (P<0.01 orP<0.05), and the fMNPCE was decreased significantly (P<0.01 orP<0.05) and tended towards normal.Conclusion Cimetidine could improve 30d survival rate of acutely irradiated mice and has good protective effect on hematopoietic system.

Objective To evaluate the safety and efficacy of percutaneous nephrolithotomy with semirigid ureteroscopic lithotripsy in semisupine-lithotomy position for treatment of encrusted ureteral stents . Methods Patients were placed in semisupine-lithotomy position .The bladder stones and ureteral encrusted stones were treated with ureteroscopy , and then percutaneous holmium laser nephrolithotomy was used to break and wash out the renal pelvis stones and renal new stones . Results The stone fragmentation and stent removal were completed in one session in all 5 cases without conversion to open surgery .The mean operative time was 65 min (range, 40-130 min).No massive hemorrhage, ureteral avulsion, or ureteral perforration occurred during the operation .Two cases had postoperative fever without sepsis .No patients had ureteral stricture or recurrent calculus within follow-ups for 3 -6 months. Conclusion Percutaneous nephrolithotomy combined with semirigid ureteroscopic lithotripsy in semisupine-lithotomy position for treatment of encrusted ureteral stents is safe and effective , being worthy of clinical promotion .

Objective To evaluate the influence of ultrasound-mediated contrast agent microbubbles carrying kallidinogenase targeted therapy on neurogenesis and angiogenesis after experimental acute cerebral infarction.Methods Kallidinogenase-loaded microbubbles were prepared using mechanical shaking method.Middle cerebral artery occlusion (MCAO) model was established in male Wistar rats by sutureoccluded method.MCAO rats (n =120) were randomly divided into 5 groups:ultrasound-mediated kallidinogenase-loaded microbubbles group (group 1),ultrasound-mediated microbubbles group (group 2),ultrasound-mediated saline group (group 3),kallidinogenase group (group 4),saline group (group 5).Medication was given through tail vein daily for 2-6 consecutive days starting 24 h after MCAO.Ultrasound groups (groups 1,2 and 3) were given 2 MHz pulse ultrasonic irradiation on the ischemia lateral skull for 10 min after injection.Cell proliferation was examined using 5'-bromo-2'-deoxyuridine (BrdU,50 mg/kg).Infarction volume and neurological function were evaluated on the 3rd,7th day after MCAO respectively.Doublecortin (DCX) + cells in the subventricular zone and laminin + in the peri-infarction region were observed at the same time.Results The number of DCX + cells in the subventricular zone (under 20 × ocular) of groups 1-5 was 251.8 ± 13.1,125.7 ± 11.6,130.2 ± 13.7,234.5 ± 12.4 and 123.7 ± 10.0 respectively.The percentage of laminin + cells in the peri-infarction region (× 10 objective) of groups 1-5 was 10.0% ± 0.8%,5.2% ± 0.7%,5.0% ± 1.0%,8.0% ± 1.8% and 5.0% ± 0.9% respectively.The number of DCX + cells in the subventricular zone and laminin + cells in the peri-infarction region of the group 1 and the group 4 was significantly more on the 7th day after MCAO compared with those of the other three groups (DCX:t values of group 1 vs groups 2,3,5 were 17.88,17.17 and 18.16,all P < 0.01 ; t values of group 4 vs groups 2,3,5 were 15.42,14.78 and 15.70,all P <0.01.Laminin:t values of group 1 vs groups 2,3,5 were 7.01,6.71 and 7.11,all P < 0.01 ; t values of group 4 vs groups 2,3,5 were 4.23,3.94 and 4.33,all P <0.01).Moreover,the number of DCX+ cells in the subventricular zone (t =2.46,P < 0.05) and laminin + cells in the peri-infarction region (t =2.78,P < 0.05) of the group 1 was much more than those of the group 4.Neurologic scores on the 7th day of groups 1-5 were 1.00 (0.75,1.25),2.0 (2.00,3.00),2.0 (1.00,2.00),1.5 (0.75,2.00),2.0 (2.00,2.50).Compared with other four groups,group 1 showed better functional improvement after stroke (U values of group 1 vs groups 2-5 were 2.0,4.0,7.5 and 2.5,all P < 0.05).While there was no significant difference in infarction volume among five groups at all the time points after MCAO.Conclusions Our study demonstrates ultrasound-mediated kallidinogenase-loaded contrast agent microbubbles targeted therapy promotes neuroblasts proliferation and vascular regeneration compared with mediated and non-medicine-loaded microbubbles therapy,which attributes to functional improvement after MCAO.Therefore,ultrasound-mediated ultrasound contrast agent microbubbles carrying drugs targeted therapy may have a perspective on ischemic stroke.

Objective:To establish the quality standard for Sipunculus nudus polysaccharide. Methods:The water content, igni-tion residues and heavy metals in Sipunculus nudus polysaccharide were determined. Authrone-sulfuric acid colorimetry was used to de-termine the polysaccharide content. Results: The water content in the polysaccharide should not exceed 5. 0%, ignition residues should not exceed 1. 0%, and the content of heavy metals should not exceed 20 ppm. The polysaccharide content should exceed 80%( glucose) . Conclusion:The method is accurate and simple, and can be effectively used in the quality control of polysaccharide in Sipunculus nudus Linnaeu.

Objective:To prepare old tea buccal tablets using wet granulation method and optimize the preparation technology. Methods:The amount of each adjuvant was studied by single factor experiments, and the formula of the buccal tablets was optimized by the orthogonal experiments using taste and disintegration time as indices. Results:The optimal formula was composed of old tea ex-tract 30 g,mannitol 60g,PEG6000 20 g,aspartame 10 g,citric acid 10 g and menthol crystal 1 g. All the tested indices including ap-pearance, hardness and disintegration time met the requirements described in Chinese pharmacopeia. Conclusion: The preparation technology is reasonable and feasible for the industrial production.

Objective To discover,diagnose and treat colorectal cancer in the early period and further improve the survival rate,reduce morbidity and mortality of the colorectal cancer by early screening for colorectal cancer in Jinjiang.Methods According to “The Guide of Cancer Screening and Early Diagnosis and Treatment in China by the Experts Group of Ministry of Health”,the case history was collected while the stool occult blood and FOBT were detected to discover high risk crowd who then inspected by electron enterscope.Results The screening was practiced in Xibin,Zimao and Neikeng town respectively.The crowd of 40-74 years old was 10 116 and 2631 of them accepted screening(acclimation rate,26%).Two hundred and fifty-seven high-risk people were discovered(9.8% of 2631)and 86 of them(acclimation rate,33%,86/257)were inspected by electron enterscope.Twenty-two cases were detected with colorectal affection(detection rate,8.6%,22/257),20 cases were diagnosed as colorectal adenoma and 2 cases were diagnosed as adenocarcinoma by pathological section.All of them accepted therapy.Conclusion Carrying out early screening for colorectal cancer is important.It can reduce morbidity and mortality of the colorectal cancer and then improve the cure rate and prolong survival in patients with colorectal cancer.

RESUMO

Sixty patients who underwent brain and neck CT angiograpby (CTA) were randomly divided into two groups: conventional protocol group and optimizing protocol group with 30 in each. The images were evaluated by two radiologists independently. The images of optimizing group avoided the artifact disturbance of the brachiocephalic vein effectively and greatly improved the imaging quality of arteria carotis communis and arteria vertebralis initial segment. Compared with the conventional group the differences were statistically significuat(P <0.05). The optimization of 64-MDCT brain and neck CTA is feasible and effective.

Nuclear factor κB (NF-κB) is an important cellular transcription factor. The important role of NF-κB-mediated cell signal transduction pathway in apoptosis is a hot topic at home and abroad. In order to discover new regulators in NF-κB signaling pathway, a high-throughput cell-based screening model based on dual luciferase reporters system was established, a number of genes that can activate NF-κB signal pathway were obtained by screening of 439 novel function genes. Among them, TMEM9B can obviously activate NF-κB signaling pathway. Further experiments showed that TMEM9B activated NF-κB signaling pathway in a dose-dependent pattern. Western blotting and EMSA experiments confirmed that TMEM9B can promote the degradation of IκBα (a cytoplasm inhibitor of NF-κB), and cause NF-κB shift from the cytoplasm to nucleus. At the same time, flow cytometry result demonstrated TMEM9B can induce apoptosis in HEK293T and HeLa cells. In short, a stable and effective screening system for NF-κB has been established, through which TMEM9B was identified to be able to significantly activate NF-κB signal transduction pathway and thus cause cells apoptosis.

METHODS: The contents of total Amino Acids and free Amino Acids in the Fruit of Eucalyptus globulus Labill. were determined with Amino Acid analyzer. RESULTS: The content of total Amino Acids in the Fruit of Eucalyptus globulus Labill. was 4. 76g, among which that of the Amino Acids essential for human body was 2. 11g ( accounting for 44. 3% ) ; the content of free Amino Acids was 25. 29mg, among which that of the Amino Acids essential for human body was 9. 3mg( accounting for 36. 7% ) . CONCLUSIONS: The Fruit of Eucalyptus globulus Labill. is rich in Amino Acids, and is of high nutritional value.

Objective To study donor liver repair in modified piggyback liver transplantation and its ~relationship to postoperative complications. Methods The relationship between donor liver repair and ~development of complications of 14 cases of modified piggyback liver transplantation,which among 17 cases of orthotopic piggyback liver allotransplantation performed in our hospital,were analyzed retrospectively.In the 14 cases,abnormal hepatic artery was repaired in 2 cases,portal vein interposition shunt was done in 1 case;~repaired ligaments were sutured in 10 of 14 cases,and not sutured in 4 cases.Results None of the 14 ~patients died in the perioperative period. The success rate of the operation was 100%. 3 patients had ~intra-abdominal bleeding,postoperatively,and in 2 cases the bleeding was related to donor liver repair. ~Postoperatively ,there was no thrombosis of hepatic artery or portal vein, and no bile duct necrosis, no hepatic outflow tract occlusion. Conclusions Donor liver repair is an important procedure in liver tramsplantation.The quality of donor liver repair is directly related to the difficulty of operative technique during liver ~transplantation ,and to the development of postoperative complications.In donor liver repair during piggyback liver transplantation,reconstruction of the retrohepatic vera cava is very important.The constructed stoma must be compatible with the donor hepatic vein outflow stoma and be able to effectively prevent the formation of eddy blood flow,ensure patency of the outflow tract and effectively prevent thrombus formation.

RESUMO

AIM: To provide a rapid method for identifying fruit of Eucalyptus globule labill. METHODS: A new method, using NIRS for rapid and nondestructive classfication of the fruit of Eucalyptus globules labill and its counterfeit drug, the fruit of Eucalyptus robusta Smith, was proposed and then cluster analysis was adopted. RESULTS: The result showed that two kinds of fruits had their own characteristic NIRS spectra. CONCLUSION: The method is quick, simple and reliable, and in combination of the fingerprint NIRS spectrum and the clustering analysis is easy to identify the adulteration.
